For me it's a three way tie between Batman, L, and Adrien Monk.

Edit: Though, if I had to pick: L
Why?
Batman=Awesome
Adrien Monk=Hilarious
L=Awesome AND Hilarious

L,BatMan(it's tough to decide who is the more interesting obsessive,mad yet brilliant detective. It says something about L that a guy in a latex Bat-Suit isn't clearly more Bat-Guano crazy than he is), Rorschach, Gillian Seed, Yuusuke Urameshi (not that he was much of a detective),Sam Spade,Phillip Lovecraft and Blacksad, Sam and Max and Professor Layton are my favorite fictional detectives or the moment.
Why arent there more videogames featuring detectives and investigative reporters, anyway?

If you're looking for that kind of thing, might I recommend the Phoenix Wright series? Sure, you technically are a defense attorney, but you act more like a detective.

We've gone this far without mention of either the Hardy Boys or Nancy Drew. I think that's kind of interesting. I was under the impression that they were very popular characters (not that I ever read either of their books).

Rorsarch, Batman, Barnaby, Rebus, Taggart, Gene Hunt, Tim Drake, The Question, most of the noir detectives I see.

In fact, I'm going to give this award to the entire noir detective genre.

Oh, and Kim Possible, because I know it's going to annoy people.
