Oh man. This thread brings back plenty of great memories :p

The first time I played Bioshock I was walking along and sucking in the atmosphere, when I stumbled upon 2 Of the first Big Daddies in the game. They were standing in a cramped room and were watching over their little sisters while staring each other down. Having barely survived my first encounter with a Big Daddy I was not about to take on 2 at once and so I strolled by with my eyes set on a ammo dispenser down the hall. I must have brushed to close to one of them because he gave me a hearty push and called me a "HNGUUHRRUH". One of the tonics I had equipped was "Electric eel" that shocks enemies when they attack you. Big Daddies apparently don't like shockwaves to the nuts so Both of them promptly started hunting me down relentlessly, drills blazing and eyes glowing red. I think my exact words were HOLY SHIT!!! o_O

5 minutes later I was barely standing upright surrounded by 2 dead big daddies, 5-6 trigger happy splicer corpses and a huge pile of thrashed security bots with the sound of 2 little sisters crying in the background :D That damn electric eel almost gave me a fucking panic attack.

I got two different ones. One from the video game world, the other from the real world!

first of all I'd have to say anywhere in the shadow temple of Ocarina of time. After that... If anybody has played a Halo 3 custom game of minotar/lambertinth..... Basically, your in a giant maze. opjective is to stay alive for more then 5-10 mins. and there's one guy powered up with a grav hammer and hunting you throughout the maze. All you can think about is "RUN RUN RUN RUN RUN!!!"

My other one from the real world...... Me and a couple guys from my school were out in a wooded area behind a elementry school shooting each other with pellet guns and aerosoft guns. It's a the middle of the night. We start going through a graveyard next to this school and all of a sudden we hear a rustle then some huge guy apparently jumpred from a tree and lands on one of my friends next to me. Me and the other guy booted it out of that graveyard just to be tackled by other huge guys. Turned out some jainitor from the elementry school called the police on us thinking we were actually shooting at each other with real guns. The huge guys who tackled us were all cops. Afterwords it was hilarious. But dam I couldn't think anything but "GET THE HELL OUT OF THERE!!! RUN RUN RUN RUN!"

I was calmly walking through the wastleland at level four. I saw something on the horizon running towards me. This is what I was thinking at the time "Is that a deathclaw? HOLY SHIT! RUN FOR YOUR LIFE!" Sadly, he caught me and pretended to be Butcher Pete.

Haaaaaaave you met Fel Reaver?
Yeah, plenty of times. The sneaky bastard always seems to come up behind me just within aggro range before I notice him, and before I even have time to say "Oh shit!" I'm but a puddle of what used to be a troll

Ohhh maan! I remember running from those ghost things in Pac-Man! It was sooo scary!

But honestly now, the biggest "OH SHIT I GOTTA RUN!" moment in game's I remember was the one in Morrowind. When I stupidly entered some cave while being only level 2 and realised that there is a shit load of monsters on the last level (there were none all the way through). When I saw one I just knew I have to run. But to be honest I was pretty scared all the way in morrowind's caves. For one reason because I was quite young then (10 I think) and because Morrowind has a real creepy atmosphere to it.
Yeah I was scarred in Morrowind I know...

Left 4 Dead on expert, doing the Death Toll campaign on co-op. Me and my friend were the only two left and about halfway through the town chapter with little health and a tank pops up. Prompt sprinting away in fear shooting and hoping before getting back to the safe room at the beginning and trying to snipe it. Needless to say that was a massive failure and resulted in us being pummelled in to the ground.

Serious Sam, when you're running away from the pyramid and something 400 feet tall with a rocket launcher for an arm breaks out of it.

The safe room wouldn't save you anyway ;). Tank can bash through the doors of safe rooms xP.

Star Wars Battlefront, when you walk right into an area where Darth Vader has decided to show up and his insane ability of deflecting any kind of lasers.
I swear he deflects any shots fired at him in that game no matter you shoot at his back or his face.
Me and my friends killed Vader with grenades. The blast pushes him. Just throw him off a cliff.

Fun times...
I actually discovered a way to kill Vader (or any hero) on Rhen Var: Harbor. In the ice caves, there is one section of a wall that doesn't have fluid dynamics, so if you walk into the wall, you fall through. I just get Vader to go flying through, usually with rockets or 'nades.

My biggest "Oh SHIT, RUN!!" moment is any time in Gears 2 when all my teammates die in the middle of wave 8 in Horde, leaving me to face Maulers, Grinders, Boomers and Cyclops with an empty Torque Bow, no grenades, and a Lancer with 3 clips left. Fun, fun.

(This happened to me last night.)

Uncharted 2 when the truck comes chasing you down in the alleyway. Definitely a memorable "oh shit!" moment.

aww man there's been so many of these, but my top two would either have to be the T-rex from the old Tomb Raider, or meeting a deathclaw for the first time in Fallout 3.
Meeting Deathclaw inner monolouge: "Wow, that's sure an ungly son-of-a-*****. I'll just take a nice little headsho-. . . Wait, why is it not exploding into bits? Oh. OOOh. OOOHHHH SHIIIIITTT!!!"

When I played Turok 2, I tranquilised this enemy standing on a ledge. When it woke up, it ran through a door and I started laughing, thinking it had run away. But a minute later i'm being attacked from behind by it! Didn't expect that at all.

Also, Half-Life 2, in Ravenholm. Ran out of bullets while standing in a shed filled with sharp things. Switched to grav gun and somehow managed to pick up the only non lethal object in the room - a bucket! Fired it at a zombies' head and ran!

Every time my units are routed in Rome: Total War, it's a literal "Oh Shit, Run!" moment.

Also, oddly enough, on Dreamfall. Later in the game, when in control of April, you have to sneak around a set of underground ruins beneath Marcuria. If a Grubber spots you they call a bigass troll monster to kill you, and as soon as that spots you (since there's no way to kill it), you pretty much have to run to a hiding place or die.

On Left 4 Dead I was running around a corner. And when I had passed that corner I came face to face with a tank who turned around and yelled. "RUUUUUUUUUUUUUUUUUUUUUN!!" I screamt in the mic. luckily though another guy was right behind me and when the tank swung I passed through that guy and his fist ended up deep in the other guys face. He saved me! :D Although he died himself..

OMG I know that scared the shit outta me and my mates the first time we played on expert. In the far end of the bridge train-car safe room in Blood Harvest we got our guns together and went outside, only to walk ten feet to hear a tank instantly. We all chuckled over our headsets and strolled back inside, casually shutting the door, to see this huge mass explode through the door and destroy us in seconds XD

the new Borderlands DLC kinda has that moment. Where you're in the bunker with the hundreds of weapon chests, and the 3 minute timer starts ticking down before the armory gets blown to hell. This is how me and my friends handled it:

*1 minute remaining*
Me: Okay, hurry it up guys, I'm full so I'm good to go any time.

Kleatus: I should be full soon

Bykov: I'm almost done.

*30 seconds*

Me: NOW GUYS, LETS GO!

Kleatus: Okay okay, i jus love lootin...

Bykov:.. done... now what?

*10 seconds*

Me: RUN!!!

Bykov: RUN WHERE?!?!

Me: I DON'T KNOW! THE NAVI JUST POINTS TO THE MIDDLE OF THE ROOM!

Kleatus:... FFFFFFFFFUUUUUUUUUUUUCC

*cutscene**armory blows up*

And then we were all a bit disappointed the stress was all for not as we were teleported back to T-Bone Junction for free and with all weapons in tow.
